A Charge Technologist from America's Respected Neighbor to the North writes: We are interested in developing a "micro" titre method (tube) in order to accommodate request for isohemagglutinin titers on pediatric (low volume) samples. Does anyone currently use such a method? The Editors are also interested in hearing from anyone who had tried it and found it to NOT work. ADDENDA Feb. 19, 2008 1. A Technical Supervisor of a hospital transfusion service in Arizona reports that her lab sometimes needs to perform isohemagglutin titers on infants for their BMT program and have developed a micro tube titer using 0.1 ml. They pipet both saline and patient plasma using MLA pipets being careful to replace the tip between tubes. Careful mixing prior to transfering from tube to tube is also important. Then they add 0.1 ml of reagent cells also using the MLA. They validated the method against traditional master tube methods and had excellent correlation. Knowing that the smaller volume is more prone to error from minor variations, they use that method for their CAP antibody titer survey and have had good results. |
